Neurons were pre-treated with 20 μM diethylumbelliferyl phosphate (DEUP), 5 μM squalene, or a mixture of DEUP and squalene and then incubated with conditioned media from 7PA2 cells (7PA2-CM) containing 10 nM Aβ42 for 2 hours. The amounts of Aβ42 in whole cell extracts, detergent-resistant membranes (DRMs) (lipid rafts), and lysosomes were measured by enzyme-linked immunosorbent assay.
