- Neurons were pre-treated with 200 nM squalestatin, 5 μM squalene, or a mixture of squalestatin and squalene and then incubated with conditioned media from 7PA2 cells (7PA2-CM) containing 10 nM Aβ42 for 2 hours. The concentrations of Aβ42 in whole cell extracts, detergent-resistant membranes (DRMs) (lipid rafts), and lysosomes were measured by enzyme-linked immunosorbent assay. Values are the mean concentration of Aβ42 ± standard deviation from triplicate experiments performed three times (n = 9). aAmounts of Aβ42 significantly lower than in control cells. bAmounts of Aβ42 significantly higher than in control cells.
